In a nutshell, a two-digit serial number prefix indicates the large frame; a three-digit prefix indicates a medium frame.

Of course,, this was mostly in reference to the early Vaquero's, due to the way Ruger marked the frames. This caused a lot of confusion to many folks. So, the easiest way to tell the differences,, was by the serial numbers, as noted above.
